《生化奇兵2》故事讲述在一代事件的10年之后，在大西洋海岸线的沿岸发生了一系列奇异事件，传说有怪物出没抓了许多小女孩潜入海底，游戏者将会进入海底城市解答这些令人疑惑的问题。而除了单人模式外，《生化奇兵2》还有多人在线联机对战模式，游戏迷将可以在网上体验海底恐怖旅程。
